Solute uptake through general porins.
General diffusion porins are among the few membrane proteins that have been thoroughly investigated by many techniques, including X-ray crystallography, AFM microscopy, computer modeling, electrophysiology and biochemistry. This had led to a good understanding of the process of solute transport per se. However, other aspects of porin function remain enigmatic, such as the molecular basis and ph...
متن کاملAntibiotic translocation through porins studied in planar lipid bilayers using parallel platforms.
In general, the method of choice to characterize the conductance properties of channel-forming bacterial porins is electrophysiology. Here, the classical method is to reconstitute single porins into planar lipid bilayers to derive functional information from the observed channel conductance. In addition to an estimated pore size, ion selectivity or transport properties in general are of importa...
متن کاملMechanisms of solute transport through outer membrane porins: burning down the house.
Porins mediate the uptake of nutrients across the outer membrane of Gram-negative bacteria. For general porins like OmpF, electrophysicoloigcal experiments now establish that the charged residues within their channels primarily modulate pore selectivity, rather than voltage-gated switching between open and closed states. Recent studies on the maltoporin, LamB, solidify the importance of its 'gr...
متن کاملThe role of Serratia marcescens porins in antibiotic resistance.
The outer membrane permeability of Serratia marcescens was studied by comparing porin-deficient mutants with their parental strains. Omp1-deficient strains were selected by moxalactam resistance, whereas mutants lacking the Omp2 porin were obtained by experimental infection with the SMP2 phage, whose primary receptor is the Omp2 porin. The role of porins was demonstrated in quinolone accumulati...
متن کاملRole of porins in intrinsic antibiotic resistance of Pseudomonas cepacia.
The measured outer membrane permeability of Pseudomonas cepacia to the beta-lactam nitrocefin was low: approximately 10 times less than that of Escherichia coli and comparable to that of Pseudomonas aeruginosa. The purified P. cepacia porin demonstrated an average single channel conductance in 1 M KCl of 0.23 nS.
